Saitama probably couldn't wield Mjolnir for the same reason Superman can't: he has the honor, humility, and desire to do good, but he doesn't see himself as a warrior. He's just an average guy who can do amazing things. Also, the fact that he only seems to fight evil when it becomes a nuisance or when he thinks it might be fun would likely get in the way of his worthiness.
Licenseless Rider might be able to though.

A lot of people keep mistaking Mjolnir as strength-based when it is more like a mechanism that only works with the correct ID signature.
If this were Marvel's Ultimate Universe, Saitama can lift Mjolnir (Hulk did).

The rules of comedy has beaten the rules of Asgardian magic. Also, he hasn't really shown interest in being a leader and would be terrible ruler of Asgard.
