No, she's asking for it to stop.
Nai is negative, but "nai ka" translates more closely to "won't you", similarly to "masen ka".
A purely direct translation of "yame nai ka?" would be "won't you stop?" Therefore, a translation of "yame na ika" would be "won't you squidly stop?"
Of course, translating directly like that isn't generally a good idea, especially in Ikamusume's case. Though, I'd have done it more like this:
"I-it squidding hurts!"
"Squidding stop it!"

True, but notice ika is in katakana, like the titles. Ika is, as I'm sure you know, is squid. But judging from her expression and the blood she wants it to stop. perhaps she hiccupped more than 30x lol.
Yeah, the way you phrased it is superior; how do you do the soft translation thing?

Click "add note" in the left panel under Options to add a generic note box to the screen.
Drag it and stretch it to the size you want, then click on the box that shows up when you hover over it to edit the text.
When you're done, click "save" to save the note to the image.
Keep in mind that like tag edits, adding notes has a 10 second cooldown to prevent abuse.
