I'm not sure what 'serious' is supposed to mean. Its a fictional creature, there are no hard rules to apply to it, unless you mean a Hollywood type project, in which case its tied to the cultural and political issues of Western views on sex and biology. If it was a real being following Wolf characteristics, then you would be able to tell if its female or male very obviously.

Not necessarily, the fur could be thick enough to hide breasts or penis, and Werewolves are often pretty jacked like a bodybuilder, so a large chest could mean anything from breasts to pectoral muscles. By serious I mean like not waifu stuff like MGE, cute stuff like goofy anime or just not campy films that don't take themselves seriously and treat werewolves as a joke.
